چکیده

Blood and lymphatic vessels in tumor tissue are major components of the tumor microenvironment. These vessels are newly formed from pre-existing host vessels stimulated by pro-blood-angiogenic and pro-lymph-angiogenic (pro-blood/lymph-angiogenic) factors expressed in tumor cells. Tumor cells establish a specific stromal microenvironment fostering tumor growth, in which blood/lymph-angiogenesis are involved. The tumor-associated blood/lymph-angiogenesis is continually induced by complicated cytokine networks, namely pro-blood/lymph-angiogenic factor-mediated paracrine and autocrine interactions among tumor cells and stromal cells including endothelial cells (ECs) and non-endothelial mesenchymal cells (neMCs). In this review, we provide an overview of the features of tumor-associated blood/lymph-angiogenesis based on recent and updated information obtained mainly from our studies. With regard to the constituent cell-dependent molecular mechanisms that regulate tumor blood/lymph-angiogenesis, we focus on: 1) the role of blood/lymph-angiogenesis-related factors/receptors expressed in tumor cells; and 2) the role of blood/lymph-angiogenesis-related factors/receptors expressed in stromal cells (ECs and neMCs). Finally, we discuss the features of tumor-associated blood/lymph-anigogenesis, especially a vessel abnormality through the viewpoint of blood/lymph-angiogenic cascades in tumor microenvironment for better understanding of the tumor vascular biology.

Suppression of prostate cancer nodal and systemic metastasis by blockade of the lymphangiogenic axis.

Lymph node involvement denotes a poor outcome for patients with prostate cancer. Our group, along with others, has shown that initial tumor cell dissemination to regional lymph nodes via lymphatics also promotes systemic metastasis in mouse models. The aim of this study was to investigate the efficacy of suppressive therapies targeting either the angiogenic or lymphangiogenic axis in inhibiting...

Collaborative interplay between FGF-2 and VEGF-C promotes lymphangiogenesis and metastasis.

Interplay between various lymphangiogenic factors in promoting lymphangiogenesis and lymphatic metastasis remains poorly understood. Here we show that FGF-2 and VEGF-C, two lymphangiogenic factors, collaboratively promote angiogenesis and lymphangiogenesis in the tumor microenvironment, leading to widespread pulmonary and lymph-node metastases. Coimplantation of dual factors in the mouse cornea...

Changing Roles of Matrix Metalloproteases and Their Inhibitors, TIMPs, During Tumor Progression and Angiogenesis

Inhibition of matrix-metalloproteinases (MMPs) by tissue inhibitors of metalloproteinases (TIMPs) has been shown in vivo to decrease metastasis and tumor-associated angiogenesis. Our laboratory is interested in understanding the role of these proteins at the pericellular microenvironment of tumor and endothelial cells. Secretion of MMPs by tumor cells enables the migration, invasion and metasta...

TIE-2-expressing monocytes are lymphangiogenic and associate specifically with lymphatics of human breast cancer

In experimental mouse models of cancer, increasingly compelling evidence point toward a contribution of tumor associated macrophages (TAM) to tumor lymphangiogenesis. Corresponding experimental observations in human cancer remain scarce although lymphatic metastasis is widely recognized as a predominant route for tumor spread. We previously showed that, in malignant tumors of untreated breast c...

Inflammation, a Key Factor in Cancer Ambush

Inflammatory condition is the consequence of defensive mechanism of immune system against viral and bacterial infection, tissue injury, UV radiation, stress and etc. Persistently acute inflammation leads to chronic phase which is characterized by production of pro-inflammatory mediators from T cells. These molecules (e.g. IL-6, TNF-&alpha, IL-1&beta and IL-17) are mostly pleiotropic cytokines i...
